    Attempts to detect retrotransposition and de novo deletion of Alus and other dispersed repeats at specific loci in the human genome

    Dispersed repeat elements contribute to genome instability by de novo insertion and unequal recombination between repeats. To study the dynamics of these processes, we have developed single DNA molecule approaches to detect de novo insertions at a single locus and Alu-mediated deletions at two different loci in human genomic DNA. Validation experiments showed these approaches could detect insertions and deletions at frequencies below 10(-6) per cell. However, bulk analysis of germline (sperm) and somatic DNA showed no evidence for genuine mutant molecules, placing an upper limit of insertion and deletion rates of 2 x 10(-7) and 3 x 10(-7), respectively, in the individuals tested. Such re-arrangements at these loci therefore occur at a rate lower than that detectable by the most sensitive methods currently available

    Background. Influenza and respiratory syncytial virus (RSV)infections cause seasonal excess mortality and hospitalisationin adults (particularly the elderly) in high-income countries. Little information exists on the impact of these infections on adults in Africa.Objectives. To estimate influenza- and RSV-related adult mortality, stratified by age and hospitalisation in Soweto.Study design. A retrospective hospital-based study in Sowetofrom 1997 to 1999 to estimate influenza- and RSV-related excess all-cause deaths and hospitalisation using a ratedifferencemethod. The study was based on influenza seasons of varying severity, provided by surveillance data.Results. Influenza seasons were significantly associated with excess mortality in adults across all 3 years, except for 18 - 64-year-olds in 1998. Excess mortality was highest in those .65 years of age: 82.8/100 000 population in the mild 1997 season and 220.9/100 000 in the severe 1998 season. Influenza significantly increased adult medical hospitalisation in the severe 1998 season alone. RSV did not significantly affect mortality or hospitalisation.Conclusion. Influenza-related mortality was substantial and disproportionately affected the elderly. Influenza vaccination for the elderly warrants consideration. The RSV-related burden was not significantly increased but merits observation over a longer period

    OBJECTIVE: To quantify normal proximal and distal oesophageal acid parameters in healthy Chinese. DESIGN: Observational study. SETTING: University teaching hospital, Hong Kong. SUBJECTS AND METHODS: Twenty healthy adults who were not on medication and were free from gastrointestinal symptoms were recruited by advertisement. Ambulatory oesophageal acid (pH5 minutes, 4/0; and the longest single acid exposure episode, 11.2/3.0 minutes. CONCLUSION: Physiological gastroesophageal reflux occurs in healthy Chinese. These initial data provide a preliminary reference range that could be utilised by laboratories studying Chinese subjects.published_or_final_versio

    Preliminary Finding: Survey on Eating Disorder and Nutritional Status among Young Athletes

    Introduction: It is well documented that eating disorders will have a negative impact on health status and athletic performance. The risks of eating disorders among athletes are required to be detected early before it affects athlete’s health and exercise performance. To date, there is still scanty information on eating disorder and nutritional status among young athletes in Sabah. Objective: The objective of this study was to determine eating disorders and nutritional status among young athletes in Sabah. Methodology: A total of 55 young athletes who are active representatives of sports competitions were selected to participate in this study. Eating attitudes test questionnaires (EAT-26) was used to determine eating disorder risk. International physical activity questionnaire (IPAQ) and 3-days food diary were used to determine nutritional status and physical activity level. Statistical Package for the Social Sciences (SPSS) version 27.0 was used to analyse the collected data. Pair-t test was used to compare energy intake of subjects with the recommended nutrient intake for Malaysia (PAL2.0). The significant level is p<0.05. Results: This study found that aesthetic and weight category sports were the highest risk for eating disorder. Energy intake (kcal/day) in the aesthetic, weight category, endurance and ball games did not significantly meet the daily energy intake recommendations. Conclusion: As a conclusion, majority of the subjects did not practice balanced diet and meet the recommended nutrient intake (RNI) especially in the aesthetic and weight category sports. These current findings are useful to detect early eating disorder among young athletes before it become critical and affect athletes’ exercise performance and health status

    Anti-inflammatory effects of indirubin derivatives on influenza A virus-infected human pulmonary microvascular endothelial cells

    Influenza A virus (IAV) poses global threats to human health. Acute respiratory distress syndrome and multi-organ dysfunction are major complications in patients with severe influenza infection. This may be explained by the recent studies which highlighted the role of the pulmonary endothelium as the center of innate immune cells recruitment and excessive pro-inflammatory cytokines production. In this report, we examined the potential immunomodulatory effects of two indirubin derivatives, indirubin-3'-(2,3-dihydroxypropyl)-oximether (E804) and indirubin-3'-oxime (E231), on IAV (H9N2) infected-human pulmonary microvascular endothelial cells (HPMECs). Infection of H9N2 on HPMECs induced a high level of chemokines and cytokines production including IP-10, RANTES, IL-6, IFN-β and IFN-γ1. Post-treatment of E804 or E231 could significantly suppress the production of these cytokines. H9N2 infection rapidly triggered the activation of innate immunity through phosphorylation of signaling molecules including mitogen-activated protein kinases (MAPKs) and signal transducer and activator of transcription (STAT) proteins. Using specific inhibitors or small-interfering RNA, we confirmed that indirubin derivatives can suppress H9N2-induced cytokines production through MAPKs and STAT3 signaling pathways. These results underscore the immunomodulatory effects of indirubin derivatives on pulmonary endothelium and its therapeutic potential on IAV-infection.published_or_final_versio

    Merkel cell carcinoma in a 17-year-old boy, report of a highly aggressive fatal case and review of the literature

    Merkel cell carcinoma is a rare tumor frequently involving the skin with an aggressive behavior and fatal outcome. It occurs mostly in the caucasian race between 60–80 years of age and it is rare in children. Herein we report our experience with a highly aggressive fatal Merkel cell carcinoma in an immunocompetent 17-year-old boy. Its characteristics and treatment modalities will be also discussed
